Tumor Slices

Tumor slices are thin, living sections of tumor tissue that preserve native architecture and interactions among cancer cells, stromal cells, and infiltrating immune cells, making them valuable models for studying disease in a near-native context. Researchers generate them by cutting freshly collected tumors into uniform sections and maintaining the slices in culture, where nutrients and oxygen diffuse through the tissue while resident cells remain spatially organized and functionally responsive. In immunology and infection research, tumor slices enable direct analysis of immune-cell activation, pathogen–tumor interactions, cytokine responses, and therapeutic effects, while reducing reliance on simplified cell lines and supporting more physiologically relevant comparisons across patient samples.

Establishing a Brain Tumor in an Organotypic Slice Co-culture Model

2025

This video demonstrates the development of a 3D co-culture system. Mouse brain slices are placed on a membrane, and fluorescently labeled human brain tumor cells are allowed to grow on it. Once the system is ready, it is stained for imaging.

A Brain Tumor/Organotypic Slice Co-culture System for Studying Tumor Microenvironment and Targeted Drug Therapies

Many types of human brain tumors are localized to specific regions within the brain and are difficult to grow in culture. This protocol addresses the role of tumor microenvironment and investigates new drug treatments by analyzing fluorescent primary brain tumor cells growing in an organotypic mouse brain slice.

Lung-tumor Tissue Explant Culture: A Procedure to Generate and Culture Precision-cut Lung Tumor Slices

2023

Organotypic primary tissue explant cultures, which include precision-cut slices, represent the three-dimensional (3-D) tissue architecture as well as the multicellular interactions of native tissue. This video describes the culturing of murine lung tumor-derived tissue slices using a rotating incubation unit, a system that enables intermittent exposure of tissues to oxygen and nutrients. The protocol describes the methodology to generate and culture precision-cut lung tumor slices for drug...

Bioluminescence Imaging to Assess Tumor Growth in an Organotypic Mouse Brain Slice

Source: Ciraku, L., et. al. An Ex Vivo Brain Slice Model to Study and Target Breast Cancer Brain Metastatic Tumor Growth. J. Vis. Exp. (2021).This video demonstrates the imaging of luciferase-expressing tumor cells in an organotypic mouse brain slice. Luciferin is added to the medium, and the plate is placed inside the chamber of the in vivo imaging system. Parameters for image capture are adjusted, and the tumor's shape and size are defined. The bioluminescent signals emitted by the tumor...
