Vero Cell Assay

A Vero cell assay is a laboratory method that uses Vero cells, a kidney-derived cell line from the African green monkey, to detect, quantify, or characterize infectious viruses. Because these cells are highly susceptible to many viruses and have limited production of type I interferons, infection can produce measurable cytopathic effects, plaques, or changes in cell viability under controlled culture conditions. Researchers use Vero cell assays to determine viral titers, evaluate neutralizing antibodies, assess antiviral activity, and support vaccine and diagnostic development. In immunology and infection research, the method provides a standardized model for examining virus-host interactions and comparing infectious samples or treatments.

Rescue of recombinant arenaviruses from cloned cDNAs, an approach referred to as reverse genetics, allows researchers to investigate the role of specific viral gene products, as well as the contribution of their different specific domains and residues, to many different aspects of the biology of arenavirus. Likewise, reverse genetics techniques in FDA-approved cell lines (Vero) for vaccine development provides novel possibilities for the generation of effective and safe vaccines to combat human...

(2016).This video demonstrates how to measure arenavirus attachment to monkey kidney cells. The assay uses low temperatures to block endocytosis and isolate the virus binding step. The approach enables precise quantification of surface-bound virus particles under conditions that restrict internalization.

Therapeutic compounds are often first examined in vitro with viability assays. Blind cell counts by a human observer can be highly sensitive to small changes in cell number but do not assess function. Computerized viability assays, as described here, can assess both structure and function in an objective manner.
