Bsa Antigen Gels

BSA antigen gels are laboratory gel systems containing bovine serum albumin (BSA), a well-characterized protein used to study antigen recognition and immune reactions. In a hydrated agar or agarose matrix, BSA and specific antibodies diffuse through the gel and form visible precipitin lines when they reach concentrations that produce antigen-antibody complexes. These gels support immunodiffusion experiments used to assess antibody specificity, compare antigenic relationships, and demonstrate principles of clinical immunology. Because BSA provides a consistent model antigen, the system also helps students and researchers evaluate assay conditions and interpret precipitation patterns relevant to diagnostic method development.

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

Development of an Antigen-driven Colitis Model to Study Presentation of Antigens by Antigen Presenting Cells to T Cells

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

A Technique for the Generation of Antigen-Specific CAR T Cells

2025

This video demonstrates a technique for generating antigen-specific chimeric antigen receptor (CAR) T cells through retroviral transduction. Activated murine CD8+ T cells are added to a microwell plate coated with recombinant fibronectin fragments. Retroviral vectors containing transgenic RNA encoding an antigen-specific chimeric antigen receptor (CAR) are then introduced, and the plate is centrifuged to facilitate transduction. After incubation, the transduced cells express antigen-specific...
