Cloned Pig Embryos

Cloned pig embryos are embryos produced by copying the nuclear genetic material of a donor pig cell, providing a model for studying development and biomedical disease. In somatic cell nuclear transfer, researchers remove the nucleus from an oocyte, insert a donor-cell nucleus, and activate the reconstructed egg so it begins dividing under controlled culture conditions. These embryos can support research on early development, genetic modification, and reproductive biology, while cloned pigs may serve as models for human diseases and potential sources of organs or tissues for transplantation. The approach also helps evaluate how nuclear reprogramming influences embryo viability and development.

Reproductive Cloning

2019

Reproductive cloning is the process of producing a genetically identical copy—a clone—of an entire organism. While clones can be produced by splitting an early embryo—similar to what happens naturally with identical twins—cloning of adult animals is usually done by a process called somatic cell nuclear transfer (SCNT). Somatic Cell Nuclear Transfer In SCNT, an egg cell is taken from an animal and its nucleus is removed, creating an enucleated egg. Then a somatic cell—any cell that is not a sex...

Molecular Cloning

Molecular cloning is a set of methods, which are used to insert recombinant DNA into a vector - a carrier of DNA molecules that will replicate recombinant DNA fragments in host organisms. The DNA fragment, which may be a gene, can be isolated from a prokaryotic or eukaryotic specimen. Following isolation of the fragment of interest, or insert, both the vector and insert must be cut with restriction enzymes and purified. The purified pieces are joined together though a technique called...

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Acclimation and Positive Reinforcement Training Pigs are social animals that respond best to calm, low-stress handling techniques. Acclimation and positive reinforcement training are important refinement strategies recommended...

All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Administration of medications, experimental drugs, and other test substances in pigs requires an appropriate route based on study objectives, compound properties, and animal welfare considerations.
