Heart Tissue Embedding

Heart tissue embedding is a histological preparation technique that supports preserved cardiac tissue so it can be cut into thin sections for microscopic examination. After fixation, the tissue is dehydrated, cleared, and infiltrated with an embedding medium, commonly paraffin or resin, which solidifies around the specimen and provides structural support during sectioning. The resulting sections can be stained to assess cardiomyocyte organization, fibrosis, inflammation, vascular changes, and other pathological features. In medicine, heart tissue embedding is essential for diagnosing cardiac disease, evaluating experimental treatments, and correlating microscopic findings with tissue injury, repair, and remodeling.

Source: Brown, A. O., et al. Visualization of Streptococcus pneumoniae within Cardiac Microlesions and Subsequent Cardiac Remodeling. J. Vis. Exp. (2015).This video demonstrates the microscopic visualization of bacterial microlesions in mouse heart tissue. The heart of a mouse infected with Streptococcus pneumoniae is harvested, washed, and fixed before being processed to obtain coronal sections. These sections are stained with hematoxylin and eosin and examined under a microscope to visualize...

This manuscript describes a soft lithography-based technique to engineer uniform arrays of three-dimensional (3D) epithelial tissues of defined geometry surrounded by extracellular matrix. This method is amenable to a wide variety of cell types and experimental contexts and allows for high-throughput screening of identical replicates.

Embedding Adult Drosophila Heads in Paraffin: Preparing Tissue for Histology

2023

This video describes how to embed Drosophila heads in paraffin using the collar method, a technique that prepares fly heads for serial sectioning. Paraffin sections are used for histological examinations of the brain's anatomy and for the assessment of neurodegeneration. The featured protocol demonstrates how multiple flies can be processed in one preparation, thereby increasing time-efficiency and minimizing experimental artifacts.
