Herbal Extract Preparation

Herbal extract preparation is the controlled process of obtaining biologically active constituents from plant material for research, formulation, or medicinal use. It typically involves selecting and processing the plant, choosing a solvent such as water or ethanol, and applying methods including maceration, infusion, percolation, or heating to transfer soluble compounds into an extract. Filtration, concentration, drying, and standardized storage help improve consistency and preserve chemical activity. In medicine, well-prepared extracts support the investigation of phytochemicals, quality control, dosage development, and evaluation of potential therapeutic effects, while careful documentation helps relate plant identity and preparation conditions to biological outcomes.

Here, we present the preparation of an er-xian decoction (EXD) in four steps—soaking, decoction, filtration, and concentration—and demonstrate the administration of a prepared EXD-containing serum to rats. These methods are applicable to the in vivo and in vitro study of herbal decoctions such as traditional Chinese medicines.

Use of Animal Model of Sepsis to Evaluate Novel Herbal Therapies

Crude and fractionated Xenopus egg extracts can be used to provide ingredients for reconstituting cellular processes for morphological and biochemical analysis. Egg lysis and differential centrifugation are used to prepare the crude extract which in turn in used to prepare fractionated extracts and light membrane preparations.

Source: Wiegand, D. J., et al. Cell-free Protein Expression Using the Rapidly Growing Bacterium Vibrio natriegens. J. Vis. Exp. (2019).This video demonstrates the preparation of a cell extract from Vibrio natriegens, a rapidly growing bacterium known for its high protein synthesis rate. The bacterial pellet is washed and resuspended in lysis buffer before sonication on ice to prevent cellular degradation. The lysate is centrifuged to separate insoluble cell debris, and the cell extract is...
