Immunomagnetic Enrichment

Immunomagnetic enrichment is a cell-separation method that isolates selected cells or particles from a mixed sample using antibodies and magnetic particles. Antibodies attached to magnetic beads bind specific surface markers on target cells, allowing a magnet to retain the labeled population while unbound cells are removed through washing or aspiration. In medicine, this approach supports the preparation of enriched cell populations for diagnostic testing, immune-cell analysis, stem-cell research, and downstream molecular assays. By improving sample purity and concentrating rare target cells, immunomagnetic enrichment can increase the sensitivity, efficiency, and reproducibility of clinical and biomedical investigations.

In this video, we demonstrate the isolation of monocytes from whole human blood by immunomagnetic negative separation. The isolated monocytes can be used for further downstream analysis.

Negative Immunomagnetic Selection: A Method to Purify B-cells from Peripheral Blood Mononuclear Cells

2023

The video explains a way to purify B-cells by immunomagnetic targeting and separation from peripheral blood mononuclear cells suspension. The method involve labeling unwanted cell types with specific antibodies or ligands (linked to magnetic particles) targeting specific cell surface proteins, to be depleted from the heterogenous cell suspension by magnetic separation.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.
