Prostate Tissue Grafting

Prostate tissue grafting is an experimental method in which prostate tissue fragments are transplanted into a recipient site to preserve and study tissue organization, growth, and function outside their original location. Grafts rely on host vascularization and continued signaling from the local microenvironment; in research models, hormone manipulation can reveal how androgen-dependent epithelium responds to changing physiological conditions. This approach supports studies of prostate development, regeneration, disease progression, and therapeutic response, while enabling researchers to examine interactions among epithelial, stromal, and immune compartments in living tissue.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.
