Thoracic Duct Cannulation

Thoracic duct cannulation is a procedure that places a catheter into the thoracic duct, the major lymphatic vessel that returns lymph from most of the body to the venous circulation. Access is typically obtained surgically or with image guidance, followed by catheter advancement within the duct to collect lymph or control its flow. The technique enables direct sampling of lymph, measurement of lymphatic transport, and administration of selected agents into the lymphatic system. In medicine and biomedical research, it supports investigation of immune trafficking, lipid absorption, and lymphatic disorders, and may assist management of chyle leakage in selected clinical settings.

Cannulation of the Mouse Submandibular Salivary Gland via the Wharton's Duct

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

This video demonstrates intraduodenal cannulation in a rat model. After anesthetizing the rat, the duodenum is located and a small puncture is created. A J-shaped cannula with a beveled tip is carefully inserted and secured, and the incision is closed. The cannulated rat model enables intra-intestinal infusion for immunopathology studies.
