U-net Cnn

A U-Net CNN is a convolutional neural network designed to assign a class or label to every pixel in an image, making it especially valuable for precise medical image segmentation. Its encoder progressively extracts contextual features while reducing spatial resolution, and its decoder restores image detail through upsampling; skip connections transfer fine-grained information from corresponding encoder layers to the decoder. In medicine, U-Net models can delineate tumors, organs, blood vessels, and other anatomical structures in imaging data such as magnetic resonance, computed tomography, and microscopy. These segmentations support quantitative analysis, diagnosis, treatment planning, and computer-assisted clinical research.

Here we present a protocol to propagate Homalodisca vitripennis cells and HoCV-1 in vitro. Medium was removed from HoCV-1 positive cultures and RNA extracted every 24 hr for 168 hr. Cell survivability was quantified by trypan blue staining. Whole virus particles were extracted post-infection. Extracted RNA was quantified by qRT-PCR.

Single Nuclei Isolation from Coronary Endarterectomy Tissue of Coronary Artery Bypass Graft Patients

Coronary endarterectomy tissue samples were processed to isolate single nuclei using an optimized dissociation protocol. The integrity of the isolated nuclei was validated through three complementary approaches: trypan blue exclusion staining to assess membrane integrity, DAPI-based confocal microscopy to confirm nuclear morphology, and flow cytometry to quantify nuclear yield and purity.

A technique for laboratory estimation of net trophic transfer efficiency of polychlorinated biphenyl (PCB) congeners to piscivorous fish from their prey is presented. To maximize applicability of the laboratory results to the field, the piscivorous fish should be fed prey fish that are typically eaten in the field.
