Bone Window Preparation

Bone window preparation is a neurosurgical technique that creates a controlled opening or thinned region in the skull to provide optical access to the brain while preserving tissue health. In rodents, researchers expose the cranial surface, carefully remove or thin bone above a target area, and often seal the site with a glass coverslip to stabilize the preparation and limit infection or mechanical disturbance. This approach supports repeated in vivo imaging, including two-photon microscopy of neuronal activity, blood flow, and neurovascular interactions, and can be combined with electrophysiology or pharmacological manipulation. Proper preparation improves experimental consistency and enables longitudinal studies of brain function and disease.
