Cerebellar Slice Preparation

Cerebellar slice preparation is a laboratory technique for isolating thin sections of cerebellar tissue so researchers can study its cellular circuitry under controlled conditions. During the procedure, tissue is rapidly removed, supported in chilled, oxygenated artificial cerebrospinal fluid, and sectioned with a vibratome to preserve neurons, synapses, and local connections. These viable slices enable patch-clamp electrophysiology, calcium imaging, pharmacological manipulation, and analysis of synaptic transmission and cerebellar network function. The preparation provides a controlled model for investigating motor learning, neuronal development, disease mechanisms, and the effects of candidate treatments.

Induction of Long-Term Depression in Cerebellar Purkinje Neurons in a Mouse Brain Slice

2025

In this video, a mouse cerebellar brain slice was used to investigate synaptic interactions between parallel fibers, climbing fibers, and Purkinje cells, which are essential for motor control. Stimulation of climbing fibers induced long-term depression (LTD) in Purkinje cells, weakening synaptic strength and resulting in reduced response to parallel fiber stimulation.
