Cortical Hemisphere Separation

Cortical hemisphere separation is a neuroscience dissection technique used to isolate the brain’s left and right cerebral hemispheres for independent examination of cortical structure and function. The procedure follows the interhemispheric fissure and carefully separates the hemispheres while minimizing damage to cortical tissue and connected structures, producing distinct samples for downstream analysis. Researchers use this preparation to compare hemispheric anatomy, gene expression, cellular organization, and neural responses, helping clarify functional lateralization and interactions between cortical regions. As a foundational tissue-preparation method, it supports studies of development, neurological disease, sensory processing, and experimental models of brain asymmetry.

We applied repetitive transcranial magnetic stimulation (rTMS) to the unilateral hemisphere of rat brain, by placing a 25-mm figure-8 coil 1 cm lateral to the vertex on the biauricular line and angulating the coil by 45°. An in-house water cooling system was used for rTMS for more than 20 min.
