Cut Loading Technique

Cut loading is a neuroscience technique that introduces fluorescent probes into neurons within freshly sectioned brain tissue, enabling cellular activity and structure to be examined in intact circuits. During the procedure, tissue is briefly exposed to a dye solution, and axons or cell processes damaged by sectioning provide entry points for otherwise membrane-impermeant indicators; the probes then diffuse through connected cellular compartments. This approach supports calcium imaging, neuronal morphology studies, and circuit analysis in acute brain slices, offering a relatively simple alternative to intracellular injection or viral labeling. It is especially useful for investigating activity patterns and connectivity across populations of neurons.

An easy and convenient method to determine the extent of gap junction tracer coupling between retinal neurons is described. This technique enables one to investigate the function of the electrical synapses between neurons in the intact retina under different illumination conditions and at different times of the day and night.

The cut-open Vaseline gap approach is used to obtain low noise recordings of ionic and gating currents from voltage-dependent ion channels expressed in Xenopus oocytes with high resolution of fast channel kinetics. With minor modification, voltage clamp fluorometry can be coupled to the cut-open oocyte protocol.
