Embryonic Kidney Cells

Embryonic kidney cells are laboratory-grown cells derived from developing renal tissue, providing a controllable system for studying cellular structure, gene expression, and signaling. In culture, researchers maintain these cells under defined conditions and introduce selected genes or molecular treatments to examine how proteins, including neuronal receptors and ion channels, are produced and function. In neuroscience, this heterologous-cell approach helps isolate the properties of individual neural molecules from the complexity of intact neurons, supporting studies of receptor pharmacology, membrane signaling, and disease-associated variants. Results can guide experiments in primary neurons and inform the development of tools or therapies targeting neural pathways.

In this report we describe a method for the isolation and culture of the progenitor cell niche from the embryonic mouse kidney that can be used to study signaling pathways regulating stem/progenitor cells of the developing kidney. These cultured cells are highly accessible to small molecule and recombinant protein treatment, and importantly also to viral transduction, which allows efficient manipulation of candidate pathways.

Source: Lucas-Hourani, M. et.al., High-throughput Screening for Broad-spectrum Chemical Inhibitors of RNA Viruses. J. Vis. Exp. (2014)This video shows a luciferase-based antiviral assay in human embryonic kidney cells infected with recombinant measles virus. It shows that serially diluted test compounds inhibit viral replication, leading to lower luminescence in a dose-dependent manner while maintaining host cell viability.
