Hindlimb Joint Scoring

Hindlimb joint scoring is a behavioral method for evaluating movement at the hip, knee, and ankle, helping quantify motor impairment and recovery after nervous system injury. During locomotor testing, an observer rates the presence, range, and coordination of joint movements according to defined criteria, converting visible hindlimb function into reproducible scores. In neuroscience, this approach is commonly used to monitor motor outcomes in animal models of spinal cord injury, neurodegeneration, and neural repair. By distinguishing changes in specific joints, hindlimb joint scoring can complement broader locomotor assessments and help determine whether experimental treatments improve functional recovery.

Surgical induction of hindlimb ischemia in the mouse is useful to examine angiogenesis, however this is compromised in certain inbred mouse strains that display marked ischemia-induced tissue necrosis. Methods are described to induce subacute limb ischemia using ameroid constrictors to circumvent this problem through the induction of gradual arterial occlusion.

We present a reproducible mouse model of heterotopic hindlimb-to-neck vascularized composite allotransplantation. This supermicrosurgical protocol, involving 0.3-0.5 mm vessel anastomoses, integrates nylon filament guidance and venous cuffing. It provides a reliable platform for translational studies on tolerance, ischemia-reperfusion injury, and immunological mechanisms in vascularized composite allotransplantation.
