Histamine Receptor Analysis

Histamine receptor analysis is the study of histamine-sensitive receptors, their distribution, signaling, and functional roles in cells and neural circuits. These receptors are G protein-coupled receptors that translate histamine binding into intracellular responses, with H1, H2, H3, and H4 receptors coupling to distinct signaling pathways; in the brain, presynaptic H3 receptors can regulate neurotransmitter release. Researchers use receptor expression profiling, ligand-binding assays, pharmacological experiments, and downstream signaling measurements to distinguish receptor subtype activity. In neuroscience, this analysis helps clarify histamine’s contributions to arousal, sleep-wake regulation, cognition, synaptic communication, and neuroinflammatory processes, while supporting the development of more selective therapeutic strategies.

Acid Suppressive Drugs for Peptic Ulcer Disease: Histamine H2-Receptor Antagonists

Histamine H2 receptors, which are intricately located on the basolateral membrane of parietal cells, play a crucial role in modulating gastric acid secretion. When released from enterochromaffin-like cells, histamine engages H2 receptors, initiating the cyclic AMP (cAMP) pathway. In this pathway, adenylyl cyclase converts ATP into cAMP, elevating intracellular cAMP levels. The activation of protein kinase A follows, stimulating the proton pump. This stimulation prompts the secretion of hydrogen...

2014

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.

Receptor trafficking modulates signaling and cell responsiveness to ligands and is, itself, responsive to cell conditions, including ligand-induced signaling. Here, we describe a powerful and flexible technique for quantitatively assessing drug-induced receptor trafficking using immunolabeling and colocalizational analysis.
