Human Rpe Cells

Human retinal pigment epithelial (RPE) cells are specialized cells that support photoreceptors and maintain the retina, making them essential for vision and a key model in neuroscience. They form a polarized monolayer between photoreceptors and the choroid, where they regulate the blood-retina barrier, recycle visual pigments, transport nutrients, and phagocytose shed photoreceptor outer segments. Studying human RPE cells helps clarify retinal development, aging, and diseases such as macular degeneration and inherited retinopathies. Primary cells and stem-cell-derived RPE models also support drug toxicity testing, disease modeling, and the development of cell-based retinal therapies.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
