Individual Fly Assay

An Individual Fly Assay is a behavioral method that measures the activity or responses of a single fruit fly under controlled laboratory conditions, helping researchers link behavior to nervous system function. By isolating one fly in a defined chamber or arena and recording its movement, orientation, or response to a stimulus over time, the assay reduces variability caused by social interactions and enables quantitative analysis. In neuroscience, individual fly assays support studies of locomotion, sensory processing, learning, and neural-circuit function, while also helping evaluate how genetic changes, environmental conditions, or experimental treatments alter behavior.

A PCR-based protocol was adapted to detect Cronobacter spp., Salmonella enterica, and Listeria monocytogenes from body surfaces and alimentary canals of individual wild-caught flies. The goal of this protocol is to detect and isolate bacterial pathogens from individual insects collected as part of an environmental sampling program during foodborne outbreak investigations.

In this video article, we describe an automated assay to measure the effect of hunger or satiety on olfactory dependent food search behavior in the adult fruit fly Drosophila melanogaster.

Drosophila Egg-Laying Preference Assay: A Method to Test Decision-Making in Flies

2023

Drosophila females choose between substrates and select sites that are attractive for egg-laying. This video describes a behavioral assay that tests their decision-making, called the egg-laying preference assay. The featured clip demonstrates the procedure with a custom-built setup, which enables researchers not only to test the behavior of many females at the same time but is also compatible with video recording and optogenetic setups.

Fly Climbing Assay: A Method to Test General Locomotion in Drosophila melanogaster

This video describes a simple method to quantify fly locomotion and shows the climbing assay performed. The fly climbing assay is a general test of motor function which relies on a negative geotaxis response.

Drosophila Aggression Assay: A Method to Test Behavioral Plasticity in Flies

2023

Aggression is an innate behavior and previous fighting experience affects fighting strategy and outcome of later contests in flies. This video describes an assay used to study Drosophila's aggressive behavior. The example protocol features the assay performed in a setup that eliminates fly handling and therefore reduces stress for the animals, which would otherwise impact the subjects' performance.
