Light Response Recording

Light response recording is a neurophysiological method for measuring how neurons or neural circuits respond to controlled illumination, making it useful for linking light stimuli to electrical activity. In a typical experiment, brief flashes or patterned light activate photosensitive cells or pathways, while electrodes or other recording systems capture changes in membrane potential, firing rate, or synaptic activity over time. These measurements help characterize response timing, sensitivity, adaptation, and circuit connectivity in visual neuroscience, supporting studies of retinal function, sensory processing, and light-driven neural modulation.

Sharp microelectrodes enable accurate electrophysiological characterization of photoreceptor and visual interneuron output in living Drosophila. Here we show how to use this method to record high-quality voltage responses of individual cells to controlled light stimulation. This method is ideal for studying neural information processing in insect compound eyes.

We developed the Visual-Motor Response to quantitate the motor output of larval zebrafish in response to light increments and decrements. We also examined zebrafish vision mutants, including the no optokinetic response (nrc) mutants, which were thought to be completely blind when tested by another vision assay, the optokinetic reflex.
