Microrna Knockdown

MicroRNA knockdown is a gene-regulation technique that reduces the activity of a specific microRNA, small noncoding RNA molecules that normally repress target gene expression. Researchers typically introduce antisense oligonucleotides, inhibitors, or microRNA sponges that bind the mature microRNA and prevent it from guiding the RNA-induced silencing complex to complementary messenger RNAs. In neuroscience, this approach helps reveal how microRNAs control neuronal differentiation, synaptic plasticity, neuroinflammation, and responses to injury. By restoring expression of affected target genes, microRNA knockdown can clarify molecular pathways involved in neurological disorders and identify potential therapeutic targets.

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.

MicroRNAs (miRNAs) are important regulators of gene expression and have been shown to play a role in numerous biological processes. To better understand miRNA-UTR interactions, we have created a genome-wide collection of 3 UTR luciferase reporters paired with a novel luciferase gene and assay reagent, the LightSwitch system.
