Mitochondria Volume Measurement

Mitochondria volume measurement is the quantitative assessment of mitochondrial size or total mitochondrial volume within cells, tissues, or neuronal compartments. It typically combines microscopy with mitochondrial labeling, image segmentation, and three-dimensional reconstruction to distinguish mitochondrial structures from surrounding cytoplasm and calculate their spatial volume. In neuroscience, these measurements help characterize mitochondrial distribution and remodeling in axons, dendrites, and synapses, where energy demand is high. Comparing mitochondrial volume across developmental stages, disease models, or experimental conditions can reveal changes in cellular bioenergetics, transport, and neuronal function, supporting research into neurodegeneration and synaptic physiology.

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.
