Nerve Gap Grafting

Nerve gap grafting is a surgical and experimental approach for repairing a peripheral nerve when injury leaves a segment missing, preventing direct reconnection of the nerve ends. A graft or nerve conduit bridges the gap, providing structural guidance and a biologically supportive pathway through which regenerating axons can extend toward their targets; autografts may also supply Schwann cells and native extracellular matrix. In neuroscience, these repairs are used to study peripheral nerve regeneration and to improve recovery after traumatic nerve injury. Research compares biological grafts, synthetic conduits, and tissue-engineered materials to restore sensory and motor function while reducing donor-site complications.

Traumatic injury to the spinal cord disrupts communication with the brain. To restore lost connectivity we utilize a peripheral nerve graft to provide a substratum for regenerating fibers in combination with neurotrophic factors and matrix-modulating enzymes to remove inhibitory molecules to promote long distance growth.

An Orthotopic Sciatic Nerve Xenograft for Neurofibromatosis Type 1 Neurofibromas

Neurofibromatosis type 1 (NF1) leads to benign peripheral nerve sheath tumors (neurofibromas) that require a specific nerve microenvironment for growth. An optimized orthotopic xenograft model was developed, using intraneural injection of human Schwann cells into NSG mice. This model recapitulates human neurofibromas and provides a valuable platform to evaluate therapeutic strategies.
