Selex Methodology

Selex Methodology is an in vitro selection method that identifies DNA or RNA aptamers, nucleic-acid ligands that bind chosen molecular targets with high affinity and specificity. By repeatedly exposing a randomized library to a target, retaining bound sequences, separating them from unbound molecules, and amplifying the enriched pool, researchers progressively favor sequences with improved binding, often applying greater selection stringency in later rounds. In neuroscience, SELEX can generate aptamers against neural proteins, receptors, or cell-surface markers for selective detection, molecular imaging, and targeted delivery. These ligands provide alternatives to antibodies and support biomarker analysis, diagnostic development, and experimental modulation of neural signaling.

In this video, we describe the systematic evolution of ligands by the exponential enrichment (SELEX) method to identify specific RNA-binding sequences for a target protein of interest. The protein is incubated with a large pool of randomized RNA sequences, and the protein-binding RNA sequences are isolated, PCR-amplified, and sequenced to identify their protein-binding sites.

The synthesis of asymmetric species of ferrocene is challenging using solution techniques. This report focuses on the methods carried out to produce a ferrocene-biotin bioconjugate using facile and clean reactions accomplished via solid-phase synthesis. Incorporation of a thiolate moiety is shown to impart the ability for immobilization on gold surfaces.

Using Luminex Corporation’s xMAP microsphere technology, we have developed the Multiplexed Fluorometric ImmunoAssay (MFIA) for serosurveillance of various laboratory animal species. The MFIA is a suspension microarray where antigen, tissue control or immunoglobulins are covalently linked to color-coded polystyrene microspheres.
