Tumor Morphology Assessment

Tumor morphology assessment is the systematic evaluation of a tumor’s cellular and tissue features to characterize its structure, behavior, and potential clinical significance. In neuroscience, researchers and pathologists examine features such as cell shape, nuclear appearance, tissue organization, vascular patterns, and the boundary between tumor and surrounding brain tissue using histological staining and microscopy. These observations help distinguish tumor types, identify features associated with malignancy, and support diagnosis and classification of central nervous system neoplasms. Morphological assessment also provides a foundation for correlating microscopic structure with symptoms, molecular findings, treatment response, and prognosis in brain tumor research and clinical care.

Source: Huang, Y. et. al., Longitudinal Morphological and Physiological Monitoring of Three-dimensional Tumor Spheroids Using Optical Coherence Tomography. J. Vis. Exp. (2019)This video demonstrates the use of optical coherence tomography (OCT) to monitor tumor spheroids in a multi-well plate by capturing high-resolution cross-sectional images. By analyzing light reflections from cellular structures, OCT constructs 3D models to assess morphological features like size and shape while also...

We describe a matrigel plug assay to illustrate angiogenic potential of a pool of factors secreted by cancer cells using two complementary imaging modalities, ultrasound and endomicroscopy. The matrigel, an extracellular matrix (ECM)-mimic gel, is utilized to introduce the host (mouse) with angiogenic factors secreted to the conditioned media...

DCE-MRI of Orthotopic Pancreatic Tumor Xenografts: A Method to Assess Microvasculature in a Target Tumor Tissue

2023

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.

Compound muscle action potential recording quantitatively assesses functional diaphragm innervation by phrenic motor neurons. Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.

A Technique to Assess the In Vivo Localization of Tumor-Specific Antibodies

2025

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.
