Shoaling Test

The shoaling test is a behavioral assay that measures shoaling, the tendency of fish to form coordinated groups with conspecifics, and provides a readout of social behavior in neuroscience research. In a typical assay, an individual fish is placed in a tank with one or more conspecifics, and investigators quantify social preference through measures such as inter-fish distance, time spent near the group, swimming alignment, and movement patterns. These responses help characterize neural circuits underlying social interaction and detect changes caused by genetic modifications, brain disorders, environmental conditions, or pharmacological treatments, including compounds that alter anxiety-like or social behavior.

This manuscript describes the setup, implementation, and analysis of boldness, aggression, and shoaling in zebrafish and testing for the presence of a behavioral syndrome. A standardized approach for behavioral quantification will allow for easier comparison across studies. Modifications to this protocol are possible as each assay can be run individually.

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.

Here, we present a protocol to use a curvelet transform-based, open-source MATLAB software tool for quantifying fibrillar collagen organization in the extracellular matrix of both normal and diseased tissues. This tool can be applied to images with collagen fibers or other types of line-like structures.

The protocol summarizes the best practices to minimize microbial bioburden in a cleanroom environment and includes strategies such as environmental monitoring, process monitoring, and product sterility testing. It is relevant for manufacturing and testing facilities that are required to meet current good tissue practice standards and current good manufacturing practice standards.
