Single Unit Recording

Single-unit recording is a neuroscience technique that measures the electrical activity of an individual neuron, allowing researchers to link cellular signals with neural function and behavior. A fine microelectrode positioned near or inside a neuron detects action potentials, or spikes, and records their timing as a spike train while experimental conditions, stimuli, or behavior are controlled. Researchers analyze firing rate, temporal patterns, and responses across conditions to study sensory processing, motor control, learning, and disease-related changes. By revealing how single neurons encode and transmit information, the method connects cellular mechanisms with activity in larger neural circuits.

Retrograde Fluorescent Labeling Allows for Targeted Extracellular Single-unit Recording from Identified Neurons In vivo

Retrograde transport of fluorescent dye labels a sub-population of neurons based on anatomical projection. Labeled axons can be visually targeted in vivo, permitting extracellular recording from identified axons. This technique facilitates recording when neurons cannot be labeled through genetic manipulation or are difficult to isolate using 'blind' in vivo approaches.

Implanting organized arrays of microwires for use in single-unit electrophysiological recordings presents a number of technical challenges. Methods for performing this technique and the equipment necessary are described. Also, the beneficial use of organized microwire arrays to record from distinct neural subregions with high spatial selectivity is discussed.
