Stem Cell Marker Analysis

Stem cell marker analysis is a set of methods used to identify and characterize stem cells by measuring molecular features associated with self-renewal, developmental potential, and lineage commitment. Researchers detect protein markers with immunocytochemistry or flow cytometry and assess marker-gene expression using methods such as quantitative PCR; comparing marker profiles helps distinguish undifferentiated cells from cells adopting neural or glial fates. In neuroscience, this analysis supports the identification of neural stem and progenitor cells, evaluation of differentiation in culture, and quality control for organoid, disease-modeling, and regenerative studies. Interpreting panels of markers rather than a single marker improves confidence in cell identity and state.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

We have identified the placenta as a major hematopoietic organ during development. We found that hematopoietic stem cells (HSCs) are both generated and expanded in the placenta in unique microenvironmental niches. Here, we describe experimental techniques required for isolation and visualization of HSCs in the mouse placenta.
