Granule Cell Layer

The granule cell layer is a densely packed neuronal layer found most prominently in the cerebellar cortex and the hippocampal dentate gyrus, where it organizes incoming signals for local circuit processing. In the cerebellum, mossy fibers excite granule cells, whose axons ascend and bifurcate into parallel fibers that distribute information across Purkinje cells; in the dentate gyrus, granule cells integrate entorhinal input and send mossy fibers to hippocampal area CA3. Studying this layer helps explain motor coordination, pattern separation, learning, and memory, while its vulnerability to altered development or excitability provides insight into neurological disease and neural circuit dysfunction.

During postnatal cerebellum development, immature granule cells originating from the germinal zone exhibit distinct modalities of migration to reach their final destination and to establish neuronal networks. This protocol describes the preparation of cerebellar slices and the confocal macroscopic approach used to investigate the factors that regulate neuronal migration.

The goal of the present protocol was to develop a method that will allow functional genomic analyses of mast cell secretion. The protocol is based on quantitative assessment of the release of a fluorescent reporter gene cotrasfected with the gene of interest and real time analyses of the secretory granule's morphology.
