Mody

Maturity-onset diabetes of the young (MODY) is a group of inherited, monogenic forms of diabetes caused by defects in pancreatic beta-cell function, making accurate diagnosis important for treatment selection. Usually transmitted in an autosomal dominant pattern, MODY results from pathogenic variants in genes that regulate insulin production, glucose sensing, or pancreatic development, producing distinct clinical phenotypes. In pharmacology, genetic subtype guides therapy: individuals with HNF1A- or HNF4A-related MODY often respond strongly to low-dose sulfonylureas, whereas other subtypes may require different approaches. Recognizing MODY can improve glycemic control, reduce unnecessary insulin use, and support family screening.

Potassium ions contribute to the resting membrane potential of cells and extracellular K+ concentration is a crucial regulator of cellular excitability. We describe how to make, calibrate and use monopolar K+-selective microelectrodes. Using such electrodes enables the measurement of electrically evoked K+ concentration dynamics in adult hippocampal slices.

This procedure describes a method for the isolation and culture of the murine organ of Corti with or without the spiral limbus and spiral ganglion neurons. We also demonstrate a method for the expression of an exogenous reporter gene in the organ of Corti explant by electroporation.

A reliable method to investigate outer hair cell (OHC) motile responses, including electromotility, slow motility and bending, is described. OHC motility is elicited by stimulation with an external alternating electrical field, and the method takes advantage of high-speed image recording, LED-based illumination, and last generation image analysis software.

Dissection of Adult Mouse Utricle and Adenovirus-mediated Supporting-cell Infection

Mechanosensory hair cells are the receptor cells of the inner ear. The best-characterized in vitro model system of mature mammalian hair cells utilizes organ cultures of utricles from adult mice. We present the dissection of the adult mouse utricle, and we demonstrate adenovirus-mediated infection of supporting cells in cultured utricles.
