Prucalopride Chronic Constipation

Prucalopride is a selective serotonin 5-HT4 receptor agonist used in pharmacology to treat chronic constipation, a disorder marked by infrequent or difficult bowel movements and delayed intestinal transit. By activating 5-HT4 receptors on enteric neurons, it enhances cholinergic signaling and coordinated propulsive contractions, helping stimulate colonic motility and accelerate transit. Its clinical use is particularly relevant when conventional laxatives provide inadequate relief, while pharmacological studies assess its effects on bowel function, tolerability, and long-term management. Understanding this mechanism illustrates how targeted modulation of gastrointestinal neurotransmission can improve symptoms without broadly stimulating the digestive tract.
