Specific Pathogen Free

Specific pathogen-free (SPF) status is a defined health designation for laboratory animals that are free from a monitored list of infectious agents, making them more consistent models for biomedical research. Animals are maintained through barrier housing, controlled breeding or rederivation, sterilized supplies, and routine microbiological screening; the designation excludes only specified pathogens, not all microorganisms. In pharmacology, SPF colonies help reduce infection-related variation in drug absorption, immune responses, toxicity, and efficacy studies. This standard improves reproducibility and supports safer interpretation of preclinical findings, while requiring ongoing surveillance because SPF status depends on the tested pathogen panel and husbandry conditions.

A Bead-Based Multiplex Immunoassay to Identify Pathogen-Specific Antibodies in Saliva

This video demonstrates a bead-based multiplex immunoassay technique to simultaneously detect antibodies for multiple environmental pathogens in human saliva. Fluorescent beads were coupled to antigens from different pathogens, which were captured using antibodies in the human saliva sample. Upon labeling the antibodies using a fluorescent reporter, the beads were analyzed using flow cytometry to assess the presence of different pathogen-specific antibodies in the saliva.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

Detection of Foodborne Bacterial Pathogens from Individual Filth Flies

A PCR-based protocol was adapted to detect Cronobacter spp., Salmonella enterica, and Listeria monocytogenes from body surfaces and alimentary canals of individual wild-caught flies. The goal of this protocol is to detect and isolate bacterial pathogens from individual insects collected as part of an environmental sampling program during foodborne outbreak investigations.
