Tnbs Model

The TNBS model is an animal model of chemically induced intestinal inflammation used to study colitis and evaluate pharmacological treatments. In this model, 2,4,6-trinitrobenzenesulfonic acid (TNBS), typically delivered into the colon with ethanol, penetrates the mucosal barrier and haptenates tissue proteins, triggering an immune response that produces mucosal injury, inflammatory-cell infiltration, and often transmural inflammation. Researchers use the model to investigate inflammatory mechanisms, assess disease severity, and test anti-inflammatory or immunomodulatory compounds. Its pathological features can resemble aspects of Crohn’s disease, making it valuable for preclinical studies while requiring careful interpretation of its chemical induction and species-specific limitations.

DNBS/TNBS induced colitis offers an alternative and inexpensive method to study the pathobiology of inflammatory bowel disease. The protocol discussed in this paper outlines the successful application of DNBS to induce colitis in mice and rats and allows one to thoroughly study host-mediated intestinal responses.

The present protocol describes the development of a Crohn's-like colitis model in rodents. Transmural inflammation leads to stenosis at the TNBS instillation site, and mechanical enlargement is observed in the segment proximal to the stenosis. These changes allow studying mechanical stress in colitis.

In this study, we describe a detailed procedure of TNBS-mediated intestinal fibrosis, which exhibits comparable pathophysiology to Crohn's fibrosis. We also discuss this approach in light of rapamycin facilitated inhibitory effects on intestinal fibrosis.
