Incidental Encoding

Incidental encoding is the formation of memories without an explicit intention to learn, making it an important process in everyday cognition and psychological research. It occurs when information is processed during another task, with attention, meaning, and the depth of processing influencing which details are represented and later retrieved. Researchers study incidental encoding to understand how environmental information enters memory, even when people are focused on a different goal. This process informs research on attention, recognition, learning, and memory retrieval, and helps explain why people may remember aspects of experiences they never deliberately tried to memorize.

Source: Laboratory of Jonathan Flombaum—Johns Hopkins University Long-term memory is a critical feature of human cognition, and it has been a prominent focus of research in experimental psychology. Many paradigms designed to tap long-term memory rely on asking participants to learn or study content, then test memory about that content. This is a good approach if one wants to understand how memory supports educational achievement, for example, where explicit study is part of the process. But,...

Information enters the brain through encoding, which is the input of information into the memory system. Once sensory information is received from the environment, the brain labels or codes it. The information is then organized with similar information and connected to existing concepts. Encoding occurs through automatic processing and effortful processing. Automatic processing involves the encoding of details like time, space, frequency, and the meaning of words, usually done without conscious...

Source: Heidbuechel, J. P., Engeland, C. E. Paramyxoviruses for Tumor-targeted Immunomodulation: Design and Evaluation Ex Vivo. J. Vis. Exp. (2019)This video demonstrates the generation of recombinant measles viruses using liposomes to deliver plasmids encoding the viral antigenome and essential helper proteins into producer cells. The transcribed and replicated RNA leads to the formation of viral particles expressing immunostimulatory and fluorescent reporter proteins. Fluorescence microscopy...

Evaluation of Immunomodulatory Activity of a Virally Encoded Bispecific T-Cell Engager

Source: Heidbuechel, J. P., et al. Paramyxoviruses for Tumor-targeted Immunomodulation: Design and Evaluation Ex Vivo. J. Vis. Exp. (2019).This video demonstrates a cell-based assay to evaluate the target-specific immunomodulatory activity of a virally encoded bispecific T-cell engager (BTE). The BTE links tumor cells expressing a target receptor with T cells, triggering tumor lysis and enzyme release. A colorimetric readout, based on substrate conversion, indicates T-cell activation. Higher...

This article will demonstrate how to monitor glutamine dynamics in live cells using FRET. Genetically encoded sensors allow real-time monitoring of biological molecules at a subcellular resolution. Experimental design, technical details of the experimental settings, and considerations for post-experimental analyses will be discussed for genetically encoded glutamine sensors.
