Protein-protein Interfaces

Muchos procesos biológicos dependen de las interacciones proteína-proteína. De hecho, un gran número de proteínas necesitan formar complejos proteicos u oligómeros para llevar a cabo sus funciones.
A veces, dos o más proteínas idénticas forman un complejo, como este dímero de kinesina, y en otros casos, diferentes proteínas o polipéptidos se unen para formar una unidad funcional. 
Por ejemplo, los microtúbulos del citoesqueleto están formados por dímeros de alfa y beta-tubulina. Las superficies de unión de los monómeros de alfa y beta-tubulina tienen formas complementarias. 
Estas formas coincidentes permiten que los monómeros formen un gran número de enlaces no covalentes entre sí, que luego mantienen unidas la tubulina alfa y beta. Este tipo de interfaz es un ejemplo de interacción superficie-superficie.

Muchas proteínas forman complejos para llevar a cabo sus funciones, lo que hace que las interacciones proteína-proteína (IBP) sean esenciales para la supervivencia de un organismo. La mayoría de los IBP se estabilizan mediante numerosas fuerzas químicas no covalentes débiles. La forma física de las interfaces determina la forma en que interactúan dos proteínas. Muchas proteínas globulares tienen formas muy parecidas en sus superficies, que forman una gran cantidad de enlaces débiles. Proteins connect with other proteins to form complexes that carry out cell functions. These protein-protein interactions, or PPIs, are important for an organism's survival. Most PPIs are held together by many weak, noncovalent chemical forces rather than one strong bond.

The shape of the interaction surface helps determine how two proteins fit together. Many globular proteins have surface shapes that match closely and can form many weak bonds. Other PPIs happen between two helices or between a surface cleft and a polypeptide chain.

Scientists study protein interfaces with both lab tests and computer methods. Laboratory tools such as affinity purification, mass spectrometry, and protein microarrays can help identify new interactions. Co-immunoprecipitation and yeast two-hybrid screening are also widely used to test whether two proteins interact in vitro.

Computer programs can predict PPIs by comparing protein sequences and three-dimensional structures with known interactions. Other methods use phylogenetic profiling, which looks for coevolving binding partners, or gene fusion analysis, which finds protein pairs fused in the genomes of other organisms. Proteins often interact with multiple partners at the same or different times, and they may contain more than one interaction interface.

Many proteins work as part of large complexes that can do their job only when all parts are present. Some interactions are also regulated, so a protein may switch partners depending on what the cell needs. Statistical and computational analysis groups these interactions into networks, and online interactome databases let users search them and help design drugs that can enhance or disrupt interactions at the interface.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
