Ligand Binding and Linkage

La mayoría de las proteínas tienen múltiples ubicaciones donde los ligandos pueden asociarse, además del sitio responsable de la función de la proteína. La unión de una molécula a cualquiera de estos sitios a menudo resulta en cambios conformacionales, alterando la forma de la proteína.
Cuando este cambio afecta a la unión de otro ligando, los dos sitios se describen como acoplados o enlazados. La unión del sitio de unión puede estar regulada positiva o negativamente.
En un enlace positivo, la unión de un ligando da lugar a cambios conformacionales que hacen que otro sitio sea más probable que se una a su ligando respectivo. 
En un ligamiento negativo, la unión de un ligando da lugar a cambios conformacionales que impiden que otro sitio de unión se asocie con su ligando.

Las proteínas alostéricas tienen más de un sitio de unión a ligando. La unión de un ligando a cualquiera de estos sitios influye en la unión de los ligandos a los otros sitios. Cuando una proteína es alostérica, sus sitios de unión se denominan acoplados o unidos. En el caso de las enzimas, el sitio que se une al sustrato se conoce como sitio activo y el otro sitio se conoce como sitio regulador. Cuando un ligando se une al sitio regulador, esto conduce a cambios conformacionales en la proteína que pueden influir en el sitio activo de la proteína.

Los enlaces de los sitios de unión pueden causar una regulación positiva o negativa de la unión del ligando a otros sitios. En los casos en los que ambos ligandos prefieren unirse a la misma conformación de una proteína, la unión en un sitio aumenta la afinidad del otro sitio por su respectivo ligando. Esto se conoce como vínculo positivo. Por otro lado, si los ligandos prefieren unirse a diferentes conformaciones, la unión de un ligando dificultará que el segundo ligando se una a la proteína. Allosteric proteins have more than one ligand binding site. A ligand is a molecule that binds to a protein. When a ligand binds at one site, it can affect binding at the other sites. These sites are called coupled or linked because they influence one another.

In enzymes, the site that binds the substrate is the active site. The other site is the regulatory site. When a ligand binds to the regulatory site, it can change the protein’s shape. This conformational change can then alter how the active site works.

Linkage between binding sites can be positive or negative. In positive linkage, both ligands prefer the same protein conformation. Binding at one site then increases the affinity, or binding strength, of the other site for its ligand. In negative linkage, the ligands prefer different conformations. Binding of one ligand makes it harder for the second ligand to bind to the protein.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism.
