Fluorometric Enzyme Assay

A fluorometric enzyme assay is a biochemical method that measures enzyme activity by monitoring fluorescence generated by a substrate or reaction product. Typically, a fluorogenic substrate is converted into a fluorescent product, and the product’s fluorescence intensity is measured at selected excitation and emission wavelengths; under controlled conditions, the signal reflects the amount of product formed and the enzyme’s catalytic rate. In biochemistry, these assays support enzyme kinetics, inhibitor screening, concentration measurements, and analysis of reaction conditions. Their high sensitivity and compatibility with microplate readers make them useful for studying low-abundance enzymes and comparing catalytic responses across samples.

High-throughput Fluorometric Measurement of Potential Soil Extracellular Enzyme Activities

To measure potential rates of soil extracellular enzyme activities, synthetic substrates that are bound to a fluorescent dye are added to soil samples. Enzyme activity is measured as the fluorescent dye is released from the substrate by an enzyme-catalyzed reaction, where higher fluorescence indicates more substrate degradation.
