Fungal Protein Extraction

Fungal protein extraction is a biochemical method for releasing and recovering proteins from fungal biomass for analysis, characterization, or downstream use. Because fungal cells have rigid chitin- and glucan-rich walls, protocols combine mechanical or enzymatic disruption with lysis buffers that solubilize proteins while limiting proteolysis; centrifugation then separates soluble extracts from cell debris. Researchers can quantify these proteins or apply fractionation and purification to examine enzymes, metabolic pathways, stress responses, and recombinant products. In biochemistry, extraction quality affects yield, protein integrity, and reproducibility, supporting proteomics, enzyme assays, and the optimization of fungal biotechnology.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.
