Cell-free Protein Synthesis

Cell-free protein synthesis is the production of proteins outside living cells using biological machinery supplied by cell extracts or purified molecular components. A DNA or RNA template directs transcription and translation in a reaction containing ribosomes, enzymes, amino acids, energy sources, and cofactors, allowing researchers to control protein production without maintaining viable cells. In bioengineering, this platform supports rapid prototyping of genetic designs, synthesis of proteins that may harm host cells, and optimization of reaction conditions. Its open, programmable format also enables studies of gene expression and the development of biosensors, diagnostics, and engineered biochemical systems.

We present the synthesis of a split-intein-mediated protein hydrogel. The building blocks of this hydrogel are two protein copolymers each containing a subunit of a trimeric protein that serves as a crosslinker and one half of a split intein.
