Cell Morphology Normalization

Cell morphology normalization is the standardization of measurements describing cell shape, size, and structure so that morphological differences can be compared reliably across samples, experiments, or imaging systems. In bioengineering workflows, researchers typically segment cells from microscopy images, extract features such as area, aspect ratio, circularity, and elongation, and normalize them against a reference population or imaging batch to reduce effects from scale, orientation, and technical variation. This approach supports quantitative assessment of cell state, phenotype, and responses to biomaterials, mechanical cues, or drug exposure. It can improve reproducibility in image-based assays and strengthen models of engineered tissues and cell-based therapies.

Here, we present a protocol for cell transplantation of zebrafish skeletal muscle and embryonal rhabdomyosarcoma (ERMS) into adult immune compromised rag2E450fs homozygous mutant zebrafish. This protocol allows for the efficient analysis of regeneration and malignant transformation of muscle cells.

Cortical networks are controlled by a small, but diverse set of inhibitory interneurons. Functional investigation of interneurons therefore requires targeted recording and rigorous identification. Described here is a combined approach involving whole-cell recordings from single or synaptically-coupled pairs of neurons with intracellular labeling, post-hoc morphological and immunocytochemical analysis.
