Dermal Epidermal Junction

The dermal epidermal junction (DEJ) is a specialized interface that connects the epidermis and dermis, providing structural support and enabling communication between these skin layers. It consists of a basement membrane and attachment structures, including hemidesmosomes and anchoring fibrils, which transfer mechanical forces and help maintain tissue organization. In bioengineering, researchers recreate this interface in engineered skin by controlling extracellular matrix composition, cell placement, and layer architecture to promote stable epidermal attachment. Accurate DEJ models support wound healing research, skin graft development, disease modeling, and evaluation of biomaterials, while improving the functional integration of laboratory-grown tissue.

Dermatitis herpetiformis (DH) is a chronic inflammatory condition characterized by an autoimmune reaction between IgA and epidermal transglutaminase (eTG). DH develops in a very small portion of gluten-sensitive and/or celiac patients. The results of this study indicate that DH can also develop in a rhesus monkey host with symptoms of idiopatic dermatitis.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

The video demonstrates the generation of neurons from adult human dermal fibroblasts (aHDFs). The aHDFs are reprogrammed using a lentiviral vector carrying genes for neuronal transcription factors and short hairpin RNAs (shRNAs).
