Genome Titer

Genome titer is the concentration of viral genome copies in a sample, providing a measure of vector or virus abundance that is important for bioengineering and gene delivery. It is typically determined by extracting nucleic acid and quantifying a target sequence with quantitative PCR against a standard curve, or by partitioning the sample into droplets for digital PCR and counting positive reactions. Genome titer supports viral vector production, batch comparison, dose normalization, and process optimization, while helping researchers distinguish physical genome abundance from functional infectivity, since genome copies do not necessarily represent biologically active particles.

Isolation and Genome Analysis of Single Virions using 'Single Virus Genomics'

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.
