Human Pancreas Tissue

Human pancreas tissue is an organ-specific biological material that combines endocrine and exocrine compartments to regulate blood glucose and support digestion, making it an important model for bioengineering research. Within the tissue, pancreatic islets release insulin and glucagon in response to changing glucose levels, while acinar and ductal cells produce and transport digestive enzymes. Bioengineers study this architecture to develop biomaterials, tissue-engineered scaffolds, organoids, and laboratory models that reproduce pancreatic structure and function. These systems support research on diabetes, drug responses, disease mechanisms, and potential cell-replacement or transplantation strategies.

Achieving high quality and appropriate quantity of human islets is one of the prominent prerequisites for successful islet transplantation. In this video, we describe step by step the procedures for human pancreatic islet isolation (part I: digestion and collection of pancreatic tissue) using a modified automated method.

The pancreas, an essential organ in the human body, is a pinkish-gray elongated structure located posterior to the stomach. It extends laterally from the duodenum towards the spleen and is firmly bound to the posterior wall of the abdominal cavity. The organ's surface has a lumpy, lobular texture that gives it a unique appearance. The broad head of the pancreas lies within the loop formed by the duodenum, while its slender body reaches towards the spleen. The tail of the pancreas is short and...

Here, we describe a method for the isolation of cells in the pancreas microenvironment from embryonic, neonatal and adult mouse tissue, focusing on the isolation of mesenchymal cells. This method allows profiling of cell gene expression and protein secretion in order to elucidate the extrinsic signals that regulate pancreas development, function, and tumorigenesis.
