Primary Renal Segments

Primary renal segments are the specialized regions of the nephron that sequentially filter, reabsorb, and secrete solutes to regulate body fluid composition and urine formation. Each segment has distinct epithelial cells, transport proteins, and permeability properties, enabling processes such as solute-driven water movement, active ion transport, and selective exchange along the nephron. In bioengineering, these functional differences guide the design of kidney-on-a-chip systems, engineered tissue models, and biomimetic filtration platforms. Reproducing segment-specific structure and transport behavior supports studies of renal physiology, drug-induced toxicity, disease mechanisms, and potential regenerative therapies.

Tissue-engineered renal constructs provide a solution for the organ shortage and deleterious effects of dialysis. Here, we describe a protocol to micro dissect murine kidneys for isolation of cortico-medullary segments. These segments are implanted into scaffold-free cellular constructs, forming renal organoids.

Early stage hemodynamic dysfunction is critical to the development of kidney disease. Yet, detection methodologies are limited. Recent advances in sonography provide a noninvasive, accurate option for early detection of kidney injury. This study outlines a step-by-step, sonographic methodology for detecting kidney dysfunction using a drug-induced nephrotoxicity rat model.
