Nonlinear Optical Imaging

Nonlinear optical imaging is a family of microscopy techniques that produces image contrast from light-induced optical responses that do not scale linearly with illumination intensity. In methods such as two-photon excitation, tightly focused pulsed lasers drive simultaneous photon absorption, while second- and third-harmonic generation convert tissue properties into signals at new wavelengths. These processes enable three-dimensional, label-free or minimally labeled imaging with optical sectioning and reduced out-of-focus excitation. In bioengineering, nonlinear optical imaging supports analysis of living cells, engineered tissues, extracellular matrix organization, and dynamic biological processes, helping researchers assess structure, function, and material behavior without extensive sample preparation.

Saturable and reverse saturable scattering were discovered in isolated plasmonic particles and adopted as a novel non-bleaching contrast method in super-resolution microscopy. Here the experimental procedures of detecting and extracting nonlinear scattering are explained in detail, as well as how to enhance resolution with the aid of saturated excitation microscopy.
