Real-time Pcr Quantification

Real-time PCR quantification is a molecular technique that measures the accumulation of amplified DNA during each polymerase chain reaction cycle, enabling sensitive analysis of target nucleic acids. Fluorescent dyes or sequence-specific probes generate signals proportional to product formation, and the cycle threshold is used with a standard curve or reference gene to estimate absolute or relative starting quantities. In bioengineering, this approach supports gene expression analysis, validation of engineered cells, pathogen detection, biomaterial studies, and evaluation of synthetic biology systems. Its rapid, quantitative readout helps researchers characterize biological constructs, monitor experimental outcomes, and optimize diagnostic or therapeutic platforms.

We describe a protocol of real time PCR to profile microRNAs in the cerebrospinal fluid (CSF). With the exception of RNA extraction protocols, the procedure can be extended to RNA extracted from other body fluids, cultured cells, or tissue specimens.
