Time-of-flight Secondary Ion Mass Spectrometry

Time-of-flight secondary ion mass spectrometry (ToF-SIMS) is a surface-sensitive analytical technique that identifies and maps chemical components by measuring ions emitted from a material, making it valuable for characterizing bioengineered systems. A focused primary ion beam bombards the sample, sputtering secondary ions from its outermost layers; these ions are separated according to their mass-to-charge ratios by measuring their flight times through a vacuum analyzer. In bioengineering, ToF-SIMS can reveal the composition and spatial distribution of biomolecules, polymers, coatings, and contaminants on biomaterials and tissue interfaces. Its high surface sensitivity supports studies of cell-material interactions, drug delivery systems, and engineered surfaces.

A two-dimensional gas chromatography-time-of-flight mass spectrometry method is described for characterization of the aqueous fraction of bio-crude produced from hydrothermal liquefaction of algae. This protocol can also be employed to analyze the aqueous fraction of liquid products from fast pyrolysis, catalytic fast pyrolysis, catalytic deoxygenation and hydro-treating.

Matrix-assisted laser desorption ionization (MALDI) is a mass spectrometry ion source ideal for the analysis of biomolecules. Instead of ionizing compounds in the gaseous state, samples are embedded in a matrix, which is struck by a laser. The matrix absorbs the majority of the energy; some of this energy is then transferred to the sample, which ionizes as a result.
