Guanosine Tetraphosphate

Guanosine tetraphosphate, commonly called ppGpp, is a bacterial signaling molecule that coordinates the stringent response, helping cells adapt to nutrient limitation and other stresses. During amino acid starvation, RelA or SpoT enzymes synthesize ppGpp from guanosine nucleotides and ATP; ppGpp then binds RNA polymerase together with the transcription factor DksA to redirect gene expression, reducing growth-related transcription while activating stress-survival pathways. Biological techniques for measuring ppGpp, including chromatography, mass spectrometry, radiolabeling, and transcriptional reporter assays, help researchers investigate bacterial physiology, antimicrobial tolerance, persistence, and host-microbe interactions. Understanding this alarmone system also supports strategies for targeting stress adaptation in pathogenic bacteria.
