Host Cell Fusion

Host cell fusion is the controlled joining of two or more living cells to form a single hybrid cell, enabling researchers to combine cellular properties and genetic material. Fusion occurs when cell membranes are brought into close contact and destabilized, commonly with polyethylene glycol, electrical pulses, or fusogenic proteins, allowing cytoplasmic mixing and sometimes subsequent nuclear fusion. This technique supports hybridoma production for monoclonal antibody generation, somatic cell genetics, and studies of viral entry and membrane biology. By creating cells with complementary traits, host cell fusion also contributes to gene transfer research, functional analysis, and the development of engineered cell systems.

Effector translocation into host cells via a type III secretion system is a common virulence strategy among gram-negative bacteria. A beta-lactamase effector fusion based assay for quantitative analysis of translocation was applied. In Yersinia infected cells, conversion of a FRET reporter by the beta-lactamase is monitored using laser scanning microscopy.
