Automated Microscopy Stage

An automated microscopy stage is a motorized platform that positions a specimen precisely beneath a microscope objective, enabling reproducible imaging across selected locations. Computer-controlled movement along the X, Y, and often Z axes coordinates with image acquisition, allowing the system to scan regions, revisit defined positions, and support automated focusing or time-lapse sequences. In biology, these stages facilitate high-throughput imaging of cells, tissues, embryos, and microbial cultures while reducing manual intervention and positional error. They are valuable for multi-position experiments, large-area imaging, longitudinal studies, and quantitative analysis of spatial or temporal changes in living and fixed samples.

We demonstrate the fabrication of a low-cost cryogenic stage designed to fit most reflected light microscopes. This lab-built cryogenic stage enables efficient and reliable correlative imaging between cryo-light and cryo-electron microscopy.

This protocol shows how to set up a standard Leginon collection and Smart Leginon Autoscreen to automate this process. This protocol can be applied to the majority of cryoEM holey foil grids.

We present an open source high content analysis (HCA) instrument utilizing automated fluorescence lifetime imaging (FLIM) for assaying protein interactions using Förster resonance energy transfer (FRET) based readouts. Data acquisition for this openFLIM-HCA instrument is controlled by software written in µManager and data analysis is undertaken in FLIMfit.

Peptide adsorption to plasticware during traditional tip-based serial dilutions can significantly impact potency determination and confound the understanding of structure-activity relationships used for lead identification and lead optimization phases of drug discovery. Here methods for automated acoustic non-contact serial dilution of peptide samples are described.

This study shows a detailed protocol to perform ultrastructure expansion microscopy in three in vitro life cycle stages of Trypanosoma cruzi, the pathogen responsible for Chagas disease. We include the optimized technique for cytoskeletal proteins and pan-proteome labeling.
